Mesenchymal Cells

چکیده
The bone marrow (BM) stroma contains a heterogeneous population of cells, including endothelial cells, fibroblasts, adipocytes and osteogenic cells, and it was initially thought to function primarily as a structural framework upon which hematopoiesis occurs. Later evidence demonstrated, however, that at least two distinct stem cell populations reside in the bone marrow of many mammalian species: hematopoietic stem cells (HSCs) and mesenchymal stem cells (MSCs), with the latter responsible for the maintenance of the non-hematopoietic bone marrow cells. MSCs, also termed multipotent marrow stromal cells or mesenchymal stromal cells, are a heterogeneous population of plastic-adherent, fibroblast-like cells, which can self-renew and differentiate into bone, adipose and cartilage tissue in culture. In the late 1960s, Friedenstein and colleagues established that single cell suspensions of BM stroma could generate colonies of adherent fibroblast-like cells in vitro. These colony-forming unitfibroblasts (CFU-Fs) were capable of osteogenic differentiation and provided the first evidence of a clonogenic precursor for cells of the bone lineage. The CFU-F assay is now widely used as a functional method to quantify stromal progenitors. Functional in vitro characterization of the stromal compartment by Dexter et al. in the 1970s then revealed its importance in regulating the proliferation, differentiation and survival of HSCs. CFU-F initiating cells in vivo have been shown to be quiescent, existing at a low frequency in human bone marrow.

Isolation and Characterization of Human Induced Pluripotent Stem Cells-Derived Mesenchymal Progenitors
Purpose: Isolating human induced pluripotent stem cells (hiPS)-derived mesenchymal progenitors as a new source of mesenchymal cells which can differentiate into different lineages like adipose and bone. Materials and Methods: After 7 days of hiPS1 culture on matrigle coated dishes, spindle like cells around colonies were removed by cell scraper. These cells that had mesenchymal like morphology ...

متن کاملP 141: Mesenchymal Stem Cells as Treatment in Neuroinflammatory Disease
Mesenchymal stem cells can be obtained from deferent tissues like adipose tissue, umbilical cord, placenta, skin, bone marrow, etc. These cells have regulatory effects on all types of immune cells such as dendritic cell, natural killers and lymphocytes. Mesenchymal stem cells induce inhibitory phenotypes of Antigen Presenting Cells (APCs) following their activity. They also change T cells pheno...

Background and Objective: Mesenchymal stem cells are promising sources of stem cells for tissue repair because of their ability to differentiate into different cells, easy proliferation and culture, and immunomodulatory properties. Despite extensive research on the immunophenotype of mesenchymal stem cells, a lack of specific markers comprises challenges for researchers. The aim of this researc...
متن کاملThe effect of bone marrow-derived mesenchymal stem cells to induce PD-L1 molecule on splenic lymphocytes
Background: Mesenchymal stem cells are non-hematopoietic stromal cells that are used in the treatment of many chronic and autoimmune diseases by modulating the immune system. Due to the limitations of using autologous mesenchymal stem cells, the use of allogeneic stem cells is a promising therapeutic approach in the treatment of immunological disorders. This study aimed to investigate the abili...
